چکیده

The shikimic acid biosynthetic pathway, which operates in micro-organisms and higher plants but not in mammals, leads to several essential amino acids and also to a variety of other metabolites. It was hoped, therefore, that blockage of the pathway might have a damaging effect on plant metabolism, giving a useful commercial herbicide with selective toxicity. Shikimate dehydrogenase, the enzyme which catalyses the conversion of dehydroshikimate (I) to shikimate (11), was chosen as the target for inhibition because it occurs early in the pathway and because of the accessibility of synthetic substrate analogues.

High-resolution structure of shikimate dehydrogenase from Thermotoga maritima reveals a tightly closed conformation.

Shikimate dehydrogenase (SDH), which catalyses the NADPH-dependent reduction of 3-dehydroshikimate to shikimate in the shikimate pathway, is an attractive target for the development of herbicides and antimicrobial agents. Structural analysis of a SDH from Thermotoga maritima encoded by the Tm0346 gene was performed to facilitate further structural comparisons between the various shikimate dehyd...

Expression, purification and properties of shikimate dehydrogenase from Mycobacterium tuberculosis.

Tuberculosis, caused by Mycobacterium tuberculosis, continues to be one of the main diseases to mankind. It is urgent to discover novel drug targets for appropriate antimicrobial agents against this human pathogen. The shikimate pathway is considered as an attractive target for the discovery of novel antibiotics for its essentiality in bacteria and absence in mammalian cells. The Mycobacterium ...

Pathway-based Screening Strategy for Multitarget Inhibitors of Diverse Proteins in Metabolic Pathways

Many virtual screening methods have been developed for identifying single-target inhibitors based on the strategy of "one-disease, one-target, one-drug". The hit rates of these methods are often low because they cannot capture the features that play key roles in the biological functions of the target protein. Furthermore, single-target inhibitors are often susceptible to drug resistance and are...

Plasmodium falciparum: interaction of shikimate analogues with antimalarial drugs.

The shikimate pathway for aromatic biosynthesis presents a target for antimalarial drug development as this pathway is absent from animals. This study extends previous work on inhibitors of the shikimate pathway, by examining their interaction with the antimalarial drugs pyrimethamine and atovaquone. Combinations of atovaquone with several shikimate analogues exhibited synergistic effects. Thes...

Inhibition and biochemical characterization of methicillin-resistant Staphylococcus aureus shikimate dehydrogenase: an in silico and kinetic study.

Methicillin-resistant Staphylococcus auerus (MRSA) strains are having a major impact worldwide, and due to their resistance to all β-lactams, an urgent need for new drugs is emerging. In this regard, the shikimate pathway is considered to be one of the metabolic features of bacteria and is absent in humans. Therefore enzymes involved in this route, such as shikimate dehydrogenase (SDH), are con...
